儿童特应性皮炎患者肠道菌群中益生菌的构成比研究

Difference of probiotics in the intestinal flora between the children with atopic dermatitis and normal controls

摘要: 目的:比较特应性皮炎(AD)患儿与正常儿童肠道菌群中益生菌的构成差异。方法:应用Illumina MiSeq测序平台对特应性皮炎患儿和健康对照儿童粪便细菌的16SrRNA基因V3-V4区进行高通量测序,比较两组益生菌属、种水平的构成差异。结果: 共纳入35例 AD 患儿和27例正常对照,在属水平,AD组患者双歧杆菌属(Bifidobacterium)物种丰度较对照组升高,而乳酸杆菌属(Lactobacillus)物种丰度较对照组降低,但二者差异均无统计学意义(P>0.05)。在种水平,AD组患者双歧杆菌属下Bifidobacterium pseudocatenulatum DSM 20438 JCM 1200 LMG 10505、unclassified_g__Bifidobacterium、Bifidobacterium_adolescentis、Bifidobacterium_animalis 物种丰度较对照组升高,而Bifidobacterium_breve物种丰度降低,差异均无统计学意义(P>0.05);AD组患者乳酸杆菌属下Lactobacillus_salivarius物种丰度较对照组降低,差异无统计学意义(P>0.05),而unclassified_g__Lactobacillus、Lactobacillus_plantarum_subsp._plantarum物种丰度升高,差异有统计学意义(P<0.05)。结论:特应性皮炎患儿肠道菌群中益生菌物种丰度与正常儿童存在差异。

Abstract: Objective: To investigate the difference of probiotics in the intestinal flora between children with atopic dermatitis (AD group) and normal children (N group). Method: The high-throughput sequencing of the 16SrRNA gene V3-V4 region of the fecal bacteria in AD group and normal control group was detected by the Illumina MiSeq sequencing platform. The difference of probiotics in the levels of the genus and species were compared. Result: Thirty five children AD and 27 normal controls were recruited. At the level of the genus, the species abundance of Bifidobacterium in AD group was higher than that in the N group, the species abundance of Lactobacillus was lower than that in N group, but there was no significant differences between them (P>0.05). At the level of the species, compared to the N group, the abundance of Bifidobacterium pseudocatenulatum_DSM_20438__JCM_1200__LMG_10505, unclassified_g__Bifidobacterium, Bifidobacterium_adolescentis and Bifidobacterium_animalis in the AD group was higher and the Bifidobacterium_breve species abundance was lower, with no significant difference (P> 0.05). The Lactobacillus_salivarius species abundance of AD group was lower than that in N group, with no significant difference (P>0.05). The species abundance of the unclassified_g_ Lactobacillus, Lactobacillus_plantarum_subsp_plantarum of AD group was higher than that in N group, with significant difference (P< 0.05). Conclusion:The abundance of probiotic species in the intestinal flora of children with atopic dermatitis is different from that of normal children.
