摘要:

目的:明确枸地氯雷他定对变应性接触性皮炎(ACD)小鼠耳廓肿胀程度的抑制作用以及对Th1/Th2相关细胞因子水平的影响。方法:建立ACD小鼠模型,观察口服不同剂量(1 mg/kg2 mg/kg3 mg/kg4 mg/kg)枸地氯雷他定对ACD小鼠耳廓肿胀的抑制作用(应先造模,后饲药物),采用ELISA 法检测小鼠血清中IL-4IFN-γ,TNF-α的水平。结果:口服各剂量枸地氯雷他定均能明显抑制ACD小鼠的耳廓肿胀程度, 且具有剂量依赖性。模型组血清IFN-γ,IL-4,TNF-α水平明显高于正常对照组(P<0.01),药物干预后血清IFN-γ,IL-4,TNF-α水平呈剂量依赖性下降。结论:枸地氯雷他定可能通过调节Th1/Th2相关细胞因子IFN-γ,IL-4,TNF-α水平对ACD小鼠的治疗发挥作用

Abstract:

Objective: To determine the effects of desloratadine citrate disodium on the edema degree of ears and the cytokine expression of Th1/Th2 cell in murine allergic contact dermatitis (ACD). Methods: The animal model of allergic contact dermatitis was established and then the inhibition effect of desloratadine citrate disodium in different doses (1 mg/kg, 2 mg/kg, 3 mg/kg and 4 mg/kg) on edema degree of ACD was assessed and the expression levels of IFN-γ, TNF-α and IL-4 were detected by ELISA. Results: Desloratadine citrate disodium can reduce the edema degree of ears. The levels of IFN-γ, TNF-α and IL-4 in serum of ACD mice were higher than that in blank controls (P<0.01) and the levels of the three cytokines in serum were all decreased after interfered by desloratadine citrate disodium, with a dose-dependent. Conclusion: Desloratadine citrate disodium is effective in the treatment of allergic contact dermatitis, which may be through regulating the levels of IFN- γ, TNF- α and IL- 4
