仙珍骨康胶囊对小鼠地塞米松骨质疏松模型关键疗效指标的影响

Effect of Xianzhengukang capsule on the therapeutic effect index in the mice model of dexamethasone osteoporosis

摘要: 目的:探索仙珍骨康胶囊防治糖皮质激素性骨质疏松症的机制。方法:40只C57BL/6小鼠随机分为地塞米松组(A组)、仙珍骨康组(B组)、地塞米松+仙珍骨康组(C组)和空白对照组(D组),每组10只。A组臀肌注射地塞米松磷酸钠,每周2次,共6周;B组予仙珍骨康混悬液20 mL/kg灌胃,每日1次;C组在A组基础上加用仙珍骨康混悬液20 mL/kg灌胃,每日1次。实验6周后,处死全部老鼠。分别采用多模式活体成像系统、透射电镜检测各组小鼠骨密度及成骨细胞形态,检测各组小鼠血清中血钙、血清无机磷(Pi)、甲状旁腺激素(PTH)、I型前胶原C-端肽(PICP)、骨钙素(BGP)、骨碱性磷酸酶(BALP)、雌二醇(E2) 、孕酮(PROG)及降钙素(CT)的水平。结果:A组骨密度与D组相比明显下降(P<0.05),C组骨密度与D组相比差异无统计学意义(P>0.05)。透射电镜检测发现A组成骨细胞溶解;B组成骨细胞结构正常;C组成骨细胞核为卵圆形,有胶原分泌;D组成骨细胞可见较多胶原分泌。 A、B、C、D组PTH水平分别为46.03±6.390017、 88.46±9.361211、67.17±5.236196, 59.71±5.552481,差异有统计学意义(P<0.05);PICP水平分别为5.59±0.839515、10.52±1.046153、 7.99±0.711197和7.38±0.72189,差异有统计学意义(P<0.05);BGP水平分别为8.59±0.813074、 4.27±0.809171、7.25±0.847148和7.77±0.753058,差异有统计学意义(均P<0.05); BALP水平分别为 2.50±0.40932、4.79±0.528068、 3.56±0.314464 和3.34±0.368841,差异有统计学意义(均P<0.05);E2水平分别为47.35±4.455982、 26.05±3.840661、38.38±3.293764和(36.42±3.326303) ,差异有统计学意义(均P<0.05); PROG水平分别为8.87±0.872026、4.37±0.79088、6.34±0.706422和6.38±0.743752,差异有统计学意义(均P<0.05);CT水平分别为27.93±3.315907、15.97±2.406497、19.61±1.820368和23.06±1.973241,差异有统计学意义(均P<0.05)。结论:仙珍骨康胶囊可能通过上调PTH、PICP、BALP,下调BGP、E2、CT与PROG水平来促进骨合成并增加骨密度。

Abstract: Objectives: To establish the mice model of dexamethasone osteoporosis and to determine the effect of Xianzhengukang capsule on the therapeutic effect index. Methods: Forty C57BL / 6 mice were divided into dexamethasone group (group A), Xianzhengukang group (group B), dexamethasone + Xianzhengukang group (group C), blank control group (group D) randomly. The mice in the group A, B, C group were respectively treated with gluteal muscle injection of dexamethasone sodium phosphate twice a week for 6 weeks, Xianzhengukang mixed suspension 20 mL/kg ,once a day, dexamethasone sodium phosphate+ Xianzhengukang. Bone mineral density and osteoblast shape were detected by multi-mode living imaging system and transmission electron microscopy (Tem).The level of blood calcium, phosphorus, parathyroid hormone (PTH), type I collagen before in mice serum C - peptide (PICP), osteocalcin (BGP), bone alkaline phosphatase (BALP), estradiol (E2), calcitonin (CT) and, progesterone (PROG) were detected. Results: The level of bone mineral density in the group A was lower than that in the group D (P<0.05) and there was no significant differences between the group C and group D (P>0.05). The osteoblast in the group A was dissolved, that in the group B was normal. The osteoblast?nuclei were ovoid and secreted collagen in the group C and D. The level of PTH in the group A, B, C were 46.03±6.390017, 88.46±9.361211, 67.17±5.236196 and 59.71±5.552481, with a significant difference (P<0.05); that of PICP were 5.59±0.839515, 10.52±1.046153, 7.99±0.711197 and 7.38±0.72189, with a significant difference (P<0.05); that of BGP were 8.59±0.813074, 4.27±0.809171, 7.25±0.847148 and 7.77±0.753058, with a significant difference (P<0.05); that of BALP were 2.50±0.40932, 4.79±0.528068, 3.56±0.314464 and 3.34±0.368841, with a significant difference (P<0.05); that of E2 were 47.35±4.455982, 26.05±3.840661, 38.38±3.293764 and 36.42±3.326303, with a significant difference (P<0.05); that of PROG were 8.87±0.872026, 4.37±0.79088, 6.34±0.706422 and 6.38±0.743752, with a significant difference (P<0.05); that of CT were 27.93±3.315907, 15.97±2.406497, 19.61±1.820368,23.06±1.973241, with a significant difference (P<0.05). Conclusions: Xianzhengukang capsule promote bone formation and increased bone mineral density, which may through up-regulating the level of PTH、PICP、BALP and down-regulating the level of BGP、E2、CT and PROG.
