Effect of Z-VAD-FMK, a broad caspase inhibitor, on the expression of cytokines released by CD4+T cell subset in the patients with atopic dermatitis

Abstract: Objective: To determine the effect of Z-VAD-FMK, a broad caspase inhibitor, on the expression of cytokines released by CD4+T cells subset in the patients with atopic dermatitis (AD). Methods: PBMCs from 30 AD patients were co-cultured with Z-VAD-FMK solution in vitro, then the cells were divided into PBS group, Z-VAD-FMK group and dexamethasone group. The levels of CD4+T cells were detected by flow cytometry. The level of IFN-γ, IL-4 and IL-17 were detected by ELISA. Results: The levels of IFN-γ, IL-4 and IL-17 in the PBS group were higher than that in the Z-VAD-FMK group (Ps<0.01) and there was no significant differences of these between the Z-VAD-FMK group and dexamethasone group (Ps>0.05). Conclusion: Z-VAD-FMK can inhibit the expression of cytokines released by CD4+T cells subset of Th1, Th2 and Th17 in patients with AD.
