Effect of RRS1 on the proliferation, metastasis and invasion of cutaneous squamous cell carcinoma cells

Abstract: Objective: To determine the effect of ribosome synthesis regulator 1 (RRS1) on the proliferation, metastasis and invasion of cutaneous squamous cell carcinoma (cSCC) cells. Methods: The difference of RRS1 expression in cSCC and normal skin was confirmed by immunohistochemical method and GEO database. SCL-1 cells with RRS1 knockdown were established by lentivirus infection. MTT, Celigo scratch, and Transwell were used to study the proliferation, metastasis and invasiveness of SCL-1 cells after RRS1 knockdown. The expression of genes related to proliferation, metastasis and invasion was detected by QRT-PCR. Results: The expression of RRS1 in cSCC cells was significantly higher than that in normal skin. The proliferation, metastasis and invasiveness of SCL-1 cells were reduced after RRS1 knockdown. After knocking down RRS1, the proliferation-related genes FGF2, AREG, and cyclin E1 were significantly reduced. Metastasis and invasion-related genes VIM, CXCL1, CXCL3, CXCL8, and MMP1 are significantly reduced. Conclusion: RRS1 may participate in the cell proliferation, metastasis and invasion of cSCC.
