Three patients with sportrichosis diagnosed by DNA sequencing

Abstract: Objectives: To report three cases with sportrichosis dignosed by DNA sequencing . Methods: DNA of tissues of three patients suspected with sportrichosis were extracted and the fungal ITS was amplified for sequencing. Pathological examination of the biopsied tissues was performed. Fungal microscopy and culture of the pus and tissue were performed. Morphology, molecular biology, sugar assimilation test and in vitro susceptibility test were performed in cultured positive colonies. Results: The molecular identification revealed S.?schenckii. The fungal culture was further identified as S. globosa. The patients were diagnosed with sportrichosis due to S. globosa. Drug sensitivity tests showed S. globosa was high sensitivity to terbinafine. The patients were treated with oral terbinafine 250 mg per day and sertaconazole cream for topical therapy. The lesions subsided after therapy. Conclusion: PCR-based DNA sequencing from infected tissues can help to identify pathogenic fungus and indicate proper antifungal treatment.
