Abstract: Primary cutaneous amyloidosis (PCA) is a chronic pruritic dermatosis characterized by deposition of amyloid protein in the dermal papillae. Conventional therapies yield unsatisfactory efficacy for refractory cases. In this study, three patients with PCA were treated with oral tofacitinib, and the efficacy and safety were evaluated via cutaneous lesion scoring and adverse event monitoring. The results demonstrated that tofacitinib rapidly relieved pruritus and continuously promoted regression of skin lesions with favourable patient tolerance and manageable adverse reactions. In conclusion, tofacitinib possesses potential clinical application value for refractory PCA.
