低氧对恶性黑素瘤A375细胞中Nodal及细胞迁移相关蛋白表达的影响

摘要: 目的:明确低氧对恶性黑素瘤A375细胞中Nodal及细胞迁移相关蛋白表达的影响。方法:体外培养A375细胞,CoCl2构建细胞内低氧模型。细胞分为空白对照组,CoCl2组,SB-431542(Nodal抑制剂)组和CoCl2 +SB-431542组。通过Western Blot技术检测Nodal、类激活素激酶4/7(ALK4/7)、与恶性肿瘤进展相关的上皮钙黏附蛋白(E-cadherin)、基质金属蛋白酶-2(MMP 2)和波形蛋白(Vimentin)的表达水平。结果:CoCl2组细胞内Nodal及ALK7蛋白浓度,高于其他组(P<0.05)。各处理组均没有检测到ALK4蛋白;CoCl2组细胞中Vimentin和MMP2蛋白的浓度高于其他组(P<0.05);各处理组E-cadherin蛋白水平差异不具统计学意义。结论:低氧环境可诱导A375细胞中Nodal和细胞迁移相关蛋白MMP2和Vimentin的高表达。

Abstract: Objective: To determine the effect of hypoxia on the expression of Nodal and the impact of relative migration protein in malignant melanoma A375 cell. Methods: Malignant melanoma A375 cells were cultured in vitro and treated with chloride (CoCl2) to construct an intracellular hypoxia model. The cells were divided into blank control group CoCl2 group, SB-431542 group and CoCl2 +SB-431542 group. The level of Nodal, Activin receptor-like kinase 4/7 (ALK4/7), E-cadherin, matrix metalloproteinase-2 (MMP-2) and vimentin was detected by Western blot. Results: The level of Nodal and ALK7 in A375 cells was higher in CoCl2 group than in other groups, with significant differences (Ps<0.05). ALK4 was not detected in all groups. The level of Vimentin and MMP2 in A375 cells was higher in CoCl2 group than in other groups, with significant differences (Ps<0.05). There was no significant difference of E-cadherin protein in different groups (P>0.05). Conclusion: Hypoxia can increase the expression of Nodal and relative migration protein of MMP2 and vimentin in the malignant melanoma A375 cells.
