胎鼠真皮成纤维细胞皮内注射对UV诱导小鼠皮肤光老化的改善作用

Improvement effect of fetal dermal fibroblasts intradermal injection on the photoaged mouse skin induced by UV

摘要: 目的:探索体外培养胎鼠真皮成纤维细胞(fibroblasts,FBs)皮内注射对小鼠背部光老化皮肤的改善作用。方法:分离培养10只第18.5天(E18.5)BALB/c胎鼠皮肤,体外培养FBs两天;用CM-Dil荧光染料进行活细胞标记,以示FBs注射后在体内存活情况。取BALB/c雄性小鼠20只,随机分为:未处理组5只、模型组5只(紫外线照射)、空白组5只(紫外线照射+注射PBS)、治疗组5只(紫外线照射+注射FBs),通过真表皮厚度及I型前胶原羧基端肽(PICP)的表达量评估胎鼠FBs对皮肤老化的改善作用。结果:胎鼠FBs形态多为梭型或不规则三角形,中央有卵圆形的细胞核,CM-Dil 细胞示踪试验显示培养细胞皮内注射后第3天活性最强。治疗组小鼠真皮厚度(243.659±6.214)μm较空白组(126.198±12.352)μm明显增厚,差异有统计学意义(P<0.05)。治疗组PICP浓度(120.357±10.260)ng/mL高于空白组(95.357±13.471)ng/mL,差异有统计学意义(P<0.05)。结论:胎鼠FBs皮内注射可改善紫外线诱导的小鼠皮肤老化。

Abstract: Objective: To investigate the effect of intradermal injection of fetal dermal fibroblasts (FBS) cultured in vitro on the photoaged dorsal skin of mice. Methods: The dermal fibroblasts were isolated and cultured from 10 BALB/c mouse embryo skins(E18.5)in vitro for two days. The living cells were labeled with CM-Dil dye to track the survival of FBs at the injection site. Twenty BALB/C male mice were randomly divided into 4 groups with 5 mice in each group: untreated group, model group (treated with ultraviolet), blank group (treated with ultraviolet +PBs injection) and treatment group (treated with ultraviolet +FBs). The effect of fetal FBs on skin photoaging was evaluated by the thickness of dermis and epidermis and the expression level of procollagen type IC-terminal propeptide (PICP). Results:  FBS of fetal mice were mostly shuttle type or irregular triangle, with oval nucleus in the center. The number of CM-Dil-labeled cells was largest on the 3rd day post-injection. The dermis thickness of the treatment group was significantly thicker than that of the blank group(243.659±6.214μm vs.126.198±12.352μm) (P<0.05). The concentration of PICP in the treatment group (120.357±10.260) ng/mL was higher than that in the blank group (95.357±13.471) ng/mL with a significant difference (P<0.05). Conclusion: Intradermal injection of fetal FBS can improve the skin photoaging induced by ultraviolet in mice.
