玉米须提取物对皮肤黑素瘤A375细胞迁移的影响及分子机制

Effects of corn silk extracts on the migration of melanoma cell line A375

摘要: 目的:明确玉米须提取物对皮肤黑素瘤迁移能力的影响。方法:培养皮肤黑素瘤细胞系A375, 0.3、3、30 μg/mL的玉米须提取物处理A375细胞,划痕法检测A375细胞的迁移能力, 实时PCR检测与迁移相关基因的表达。结果:玉米须提取物组A375细胞迁移能力低于空白对照组,且浓度越高迁移力越弱;玉米须提取物组上皮间质转化(EMT)相关基因TWIST1、SNAI1、SNAI2、VIM的表达量低于空白对照组, PTGS2基因表达量高于对照组。结论:玉米须提取物能抑制黑素瘤A375细胞迁移能力,下调EMT相关基因表达。

Abstract:

Objective: To determine the effect and mechanisms of corn silk extracts on the migration of cutaneous melanoma. Methods: The skin melanoma cell line A375 was cultured and was treated by corn silk extracts in the concentration of 0.3, 3,30 μg/mL. The migration of A375 cells was detected by scratch-wound assay and the expression of migration related genes was detected by the Real time PCR. Results: The migration number of A375 cells in the group of corn silk extracts was less than that in the blank group and the higher the concentration, the less migration number was. Compared to the blank group, the expression level of EMT associated genes (TWIST1, SNAI1, SNAI2 and VIM) was lower and the expression level of PTGS2 was higher in the group of corn silk extracts. Conclusion: The corn silk extracts can inhibit the migration of the skin melanoma and increase the expression of EMT related genes.
