Effect of 18β-glycyrrhetinic acid new derivatives on proliferation of HaCaT cells

Abstract: Objective: To determine the effect of 18β-glycyrrhetic acid new derivative 3-O-(5-methylpyrazine-2-formyl)-glycyrrhetinic acid (GA2) on the proliferation of HaCaT cells. Methods: HaCaT cells were cultured in vitro, and then, the cells were divided into the blank control group, GA2 groups with different concentrations of (31.2, 62.5, 125 μM). The proliferation of HaCaT cells was detected by CCK8, EdU method, cell cloning, and flow cytometry. The Caspase 3 expression level was detected through Caspase 3 activity. Results: After HaCaT cells treated with 31.2, 62.5 and 125 μM of GA2 for 24 hours, the cell inhibition rates and apoptosis rates were increased in a dose-dependent manner. The cell cloning and cell proliferation ability were decreased in a dose-dependent manner. The expression level of Caspase 3 was increased in a dose-dependent manner. Conclusion: GA2 can inhibit the proliferation of HaCaT cells, which may be related to the activating Caspase 3.
