Longdanxiegan decoction activates Bax/Caspase-3 pathway and induces the apoptosis of HaCaT cells

Abstract: Objective: To determine the role of Longdanxiegan decoction in the apoptosis of HaCaT cells. Methods: The effect of Longdanxiegan decoction on the apoptosis of HaCaT cells was detected by Tunel method. The apoptosis of HaCaT cells was detected by fluorescence microscopy after DAPI staining. The effect of HaCaT cells on the expression of apoptosis-related proteins caspase-3, Bcl-2 and Bax was detected by Western blot. Results: The abnormal cells shape rate and the apoptosis rate were higher in the Longdanxiegan decoction group than in the untreated group. Longdanxiegan decoction significantly down-regulated the expression level of Bcl-2 and up-regulated Bax, and actived caspase in HaCaT cells. The cell apoptosis caused by Longdanxiegan decoction decreased in the cells pretreated by Z-DEVD-FMK (inhibitor of caspase-3). Conclusion: Longdanxiegan decoction can significantly induce apoptosis of human keratinocytes, which may be through down-regulation of Bcl-2 and up-regulation of caspase-3 or Bax.
