龙胆泻肝汤激活Bax/Caspase-3通路并诱导人角质形成细胞凋亡

Longdanxiegan decoction activates Bax/Caspase-3 pathway and induces the apoptosis of HaCaT cells

摘要: 目的:明确龙胆泻肝汤对人角质形成细胞株HaCaT细胞凋亡的影响。方法:Tunel法检测龙胆泻肝汤对HaCaT细胞凋亡的影响; DAPI染色后荧光显微镜观察HaCaT细胞凋亡形态变化; Western blot检测分析HaCaT细胞凋亡相关蛋白表达。结果:龙胆泻肝汤处理组HaCaT细胞形态发生了明显变化,凋亡比例明显高于未处理细胞组。龙胆泻肝汤作用于HaCaT细胞后活化的caspase-3和Bax的表达明显上调, Bcl-2蛋白表达下降。caspase-3特异性抑制剂Z-DEVD-FMK预处理后可降低龙胆泻肝汤诱导的细胞凋亡。结论:龙胆泻肝汤能明显诱导人角质形成细胞的凋亡,其机制可能与下调Bcl-2的表达,增高Bax和caspase-3的表达有关。

Abstract: Objective: To determine the role of Longdanxiegan decoction in the apoptosis of HaCaT cells. Methods: The effect of Longdanxiegan decoction on the apoptosis of HaCaT cells was detected by Tunel method. The apoptosis of HaCaT cells was detected by fluorescence microscopy after DAPI staining. The effect of HaCaT cells on the expression of apoptosis-related proteins caspase-3, Bcl-2 and Bax was detected by Western blot. Results: The abnormal cells shape rate and the apoptosis rate were higher in the Longdanxiegan decoction group than in the untreated group. Longdanxiegan decoction significantly down-regulated the expression level of Bcl-2 and up-regulated Bax, and actived caspase in HaCaT cells. The cell apoptosis caused by Longdanxiegan decoction decreased in the cells pretreated by Z-DEVD-FMK (inhibitor of caspase-3). Conclusion: Longdanxiegan decoction can significantly induce apoptosis of human keratinocytes, which may be through down-regulation of Bcl-2 and up-regulation of caspase-3 or Bax.
