两株非典型烟曲霉临床株抗真菌药物敏感性和形态异常相关分子机制研究

Antifungal sensitivity and relevant molecular mechanisms of two atypical Aspergillus fumigatus

摘要: 目的:比较两株白色不产孢烟曲霉临床株(A1j和A2j)的抗真菌药物敏感性及形态异常产孢基因与Af293的差异性。方法:采用改良的琼脂稀释药敏实验方法检测A1j和A2j与模式对照菌Af293对伏立康唑(VRC)、 伊曲康唑(ITC)和两性霉素B(AMB)的敏感性。采用PCR和RT-PCR的方法对A1j和A2j产孢相关基因brlA和wetA进行检测。结果:抗真菌试剂可抑制Af293、A2j和A1j的生长,差异无统计学意义。产孢诱导6h后,Af293中brlA基因转录水平显著上升,而A1j和A2j中对应的产孢基因转录水平无明显变化。brlA基因测序结果示A1j菌株无突变,A2j存在TCCT碱基缺失。结论:非典型烟曲霉A1j和A2j与标准株Af293在抗真菌药物敏感性方面没有差异,A2j存在形态异常产孢基因brlA突变。

Abstract: Objective: To compare the difference of in antifungal drug sensitivity and morphological related molecular mechanisms between the control strain Af293 and the two newly  clinical strain of atypical Aspergillus fumigatus (A1j and A2j). Methods: The antifungal susceptibility of A2j, A1j and Af293 to amphotericin B (AMB), itraconazole (ITC) and voriconazole (VRC) was detected by modified agar dilution susceptibility test. The method of was used to study the genes related to sporulation (brlA and wetA) were detected in A1j and A2j by RT-PCR and PCR. Results: Amphotericin B (AMB), itraconazole (ITC) and voriconazole (VRC) could inhibite the bacterial colonies of A1j, A2j and Af293, with no significant difference. After spores were induced for 6 hours, the expression level of brlA in Af293 strain was increased, but that of brlA and wetA in A1j and A2j strain was no change. The sequencing found a mutation of 597_600delTCCT of brlA in A2j and no mutation was found in A1j. Conclusion: There was no significant difference of the antifungal susceptibility of A2j, A1j and Af293 to antifungal drug. There was a mutation in sporulation gene brlA related to morphological abnormality of strain A2j.
