摘要: 目的: 明确杨梅黄酮对人皮肤成纤维细胞增殖及胶原合成的影响。方法: 原代培养的人皮肤成纤维细胞加入不同浓度(0,10,25,50,75,100μM)的杨梅黄酮,流式细胞仪检测细胞周期,MTT检测细胞增殖活性和胶原合成。结果: 杨梅黄酮浓度为10μM及25μM时成纤维细胞增殖活性明显增加;浓度高于50μM处理后的成纤维细胞增殖活性降低,I型胶原表达减少。结论: 杨梅黄酮大于50μM时能抑制成纤维细胞胶原合成,提示该药具有较好的抗皮肤和内脏器官纤维化作用,为治疗纤维化相关疾病提供了一定的理论依据。

Abstract: Object To determine the effect of myricetin on cell proliferation and collagen synthesis of human fibroblast. Methods: The primary cultured fibroblasts in vitro were isolated and treated with myricetin in different concentration (0, 10, 25, 50, 75, 100μM). Cell cycle was measured by flow cytometry. The fibroblasts proliferation and the expression of collagen type I was assessed by MTT assay. Results: The cellular viability of fibroblast was increased in the dose of 10 μM and 25 μM of myricetin, and was decreased in the dose of 50μM or above. Conclusions: The proliferation and collagen synthesis of human fibroblast can be inhibited when the concentration of myricetin is higher than 50μM, which provides a theoretical base for the treatment of fibrosis diseases.
