肠道菌群代谢产物IAID对特应性皮炎小鼠模型的作用

The impact of gut microbial metabolite on mouse model of atopic dermatitis

摘要: 目的:确定肠道菌群代谢产物吲哚-3-甲醛(IAID,indole-3-carboxaldehyde)在特应性皮炎(AD)小鼠模型中的作用。方法:将C57小鼠随机分为对照组、模型组和IAID组。小鼠双耳外涂卡泊三醇(MC903)制备AD模型,IAID组给予65 μg/kg IAID灌胃,模型组灌等体积DMSO作为对照,对照组不处理。10天后肉眼观察大体炎症情况,千分尺测量小鼠耳廓厚度,ELISA法检测小鼠血清中总IgE水平并通过RT-PCR的方法检测全皮中IL4,IL5,IL6,IL13,IL33,TSLP的表达。结果:IAID组小鼠与模型组相比大体炎症明显减轻,血清总IgE水平明显降低(P<0.001),皮损IL13、TSLP的mRNA水平也显著下降(P<0.05)。结论:肠道菌群代谢产物IAID可以缓解AD小鼠模型的皮肤炎症。

Abstract: Objective: To determine the impact of gut microbial metabolite indole-3-carboxaldehyde (IAID) on mouse model of atopic dermatitis (AD). Methods: C57 mice were divided into 3 groups: control group, model group and IAID group. Vitamin D3 analog MC903 was applied on on mice ear to establish an AD-like model. The mice in IAID group were gavaged with 65 μg/kg IAID, and the same amount of DMSO as a vehicle control through administration by gavaging. At day 10, The mice ears were observed for any inflammatory sign and the thickness of the ears were measured. The RT-PCR technique was used to detect the expression of IL4, IL5, IL6, IL13, IL33, TSLP in whole skin and ELISA technique was used to detect the level of total IgE. Results: Compared to model group, we observed a obvious alleviation in IAID group with a decreased level of total IgE (P<0.001) and IL13, TSLP (P<0.05). Conclusion: Gut microbial metabolite IAID can relieve the severity of AD-like inflammation in the mice model.
