染料木素对UVA诱导成纤维细胞急性光损伤的防护作用

摘要: 目的:明确染料木素对UVA诱导成纤维细胞急性光损伤的防护作用。方法:将体外培养的成纤维细胞分为空白对照组,UVA照射对照组及UVA+染料木素(0.01,0.1,1,10,100umol/L)组。采用CCK8法测定细胞增殖情况,流式细胞术检测细胞凋亡情况, RT-PCR检测Sirt1mRNA的表达水平。结果:与UVA组比较,UVA+0.01,0.1,1,10 μmol/L染料木素组细胞增值活性及Sirt1mRNA表达明显升高,细胞凋亡率降低(均P<0.05);而UVA+100μmol/L染料木素,细胞凋亡率无明显差异(P>0.05);结论:0.01~10μmol/L染料木素对UVA诱导成纤维细胞的急性光损伤有防护作用。

Abstract: Objective: To determine the protective effect of genistein on acute light damage induced by UVA in fibroblasts cells. Methods: Fibroblasts cells cultured in vitro were divided into the control group, UVA group and UVA +0.01,0.1,1,10,100 μmol/L genistein group. The proliferation activity, apoptosis rate and the level of Sirt1mRNA were detected by the cck8 experiment method, flow cytometry and RT-PCR, respectively. Results: Compared with the UVA group, the proliferation activity and the level of Sirt1mRNA in the UVA+ 0.01,0.1,1,10μmol/L genistein group was significantly increased and the apoptosis rate was significantly decreased (Ps<0.05), while there was no significant difference in apoptosis rate between the UVA +100 umol/L genistein group and UVA group (P>0.05). Conclusion: 0.01-10 μmol/L genistein is effective in protecting the fibroblasts cells from acute light damage induced by UVA.
