NB-UVB对黑素细胞黑素合成相关基因表达的影响

摘要: 目的:研究NB-UVB照射对黑素细胞的细胞活性、黑素合成、黑素合成相关基因表达的影响。方法:培养黑素细胞系PIG1,利用MTT法检测不同剂量NB-UVB(400、800、1200、1600、2000 mJ/cm2)照射后细胞的增殖率,采用NaOH裂解法测定细胞中的黑素含量,RT-PCR和Western blot方法检测UVB照射后12个黑素合成相关基因Tyr、Tyrp1、Tyrp2、OA1、MART-1、Pmel17、VAT-1、OCSP、syntenin、CHCHD3、flotillin-1、GPNMB的mRNA和蛋白的表达。结果:NB-UVB照射剂量在400、800、1200 mJ/cm2 时,细胞活力无明显变化,照射剂量为1600、2000 mJ/cm2时细胞活力明显下降。NB-UVB照射剂量为400、800、1200 mJ/cm2 时,随着照射剂量的增加,黑素的合成增加;NB-UVB照射后,12种黑素体蛋白中,Tyr、Tyrp2、GPNMB、Syntenin、MART-1的基因mRNA及蛋白表达显著升高。结论:UVB照射显著增加了黑素合成和相关基因的表达。

Abstract: Objective: To study the effects of NB-UVB irradiation on the melanocytes viability, melanin synthesis, and melanin synthesis-related gene expression. Methods: The melanocyte cell line PIG1 was cultured. The cell proliferation after different doses of UVB irradiation (400, 800, 1200, 1600, 2000 mJ/cm2) was detected by MTT method. The melanin content was detected by NaOH lysis method. The expression of melanin synthesis-related genes, Tyr, Tyrp1, Tyrp2, OA1, MART-1, Pmel17, VAT-1, OCSP, syntenin, CHCHD3, flotillin-1, GPNMB after UVB irradiation were detected by RT-PCR and Western blot. Results: The cell viability decreased significantly after irradiated by NB-UVB radiation at dose of 1600 and 2000 mJ/cm2. There was no significant change of cell viability after irradiated by NB-UVB radiation at dose of 400, 800 and 1200 mJ/cm2, and melanin synthesis increased with increasing radiation dose. After ultraviolet irradiation, of the 12 melanin synthesis-related genes, 5 of Tyr, Tyrp2, GPNMB, Syntenin, MART-1 mRNA and protein were significantly increased. Conclusion: NB-UVB irradiation significantly increased the melanin synthesis and the expression of related genes.
