GPR78与ETS1基因交互作用与系统性红斑狼疮的相关性研究

Interactions between GPR78 and ETS1 gene associate with systemic lupus erythematosus

摘要:

目的:明确GPR78ETS1基因的交互作用对系统性红斑狼疮(SLE)发病的影响。方法:在前期GWAS的基础上,选择可能存在共同生物学效应的GPR78SNP rs3103074)和ETS1SNP rs6590330),在中国汉族人群4199SLE8255名对照中基因分型。通过logistic回归模式检验这两者之间可能存在的交互作用。结果:GPR78ETS1基因在中国汉族人SLE发病中存在显著的交互作用(Pcombined=1.4×10-4,OR=1.27)。单个SNPSLE的关联性分析发现GPR78SNP rs3103074)是提示点(P=6.2×10-4,OR=1.22,95% CI:1.12~1.32),与SLE中度关联。结论:GPR78ETS1基因之间存在着显著的交互作用,可能在SLE的发病中发挥作用。

Abstract:

Objective: To determine the potential effects of the interaction of GPR78 (rs3103074) and ETS1 (rs6590330) on systemic lupus erythematosus (SLE). Methods: Based on our previous GWAS of SLE, GPR78 (SNP rs3103074) and ETS1 (SNP rs6590330) (maybe have an interaction in the pathogenesis of SLE) were chosen. In total, 4199 patients and 8255 controls from Chinese Han population were genotyped for GPR78 rs3103074 and ETS1 rs6590330. Pair-wise interactions between SNPs were evaluated from likelihood ratio tests utilizing logistic regression models. Results: A significant interactive effect between GPR78 rs3103074 and ETS1 rs6590330 on SLE susceptibility (Pcombined=1.4×10-4,OR=1.27) was found through logistic regression analysis. GPR78 rs3103074 was suggestive of SNP, showed medium association with SLE (Pcombined=6.2×10-4,OR=1.22, 95% CI:1.12-1.32) through single SNP association analyses. Conclusion: There is a significant interaction between gene GPR78 and ETS1, which may play a role in the pathogenesis of SLE.
