Expression of interleukin-35 in atopic dermatitis-like murine model

Abstract: Objective: To detect the expression of IL-35 in atopic dermatitis-like murine model. Methods: Thirty BALB/c mice were randomly divided into the model group and control group (15 in each). The atopic dermatitis-like models were established by topical application of 0.5% 2,4-dinitrofluorobenzene. After successful modeling, the serum levels of IL-35, IL-4, IFN-γ and IL-17 were detected by ELISA and the expressions of IL-12p35 and EBI3 mRNA were tested by RT-PCR. Results: In the model group, the inflammation was obvious in the skin of the back The histopathology of skin lesions showed thickness of the epidermis, spongiosis and infiltration of inflammatory cells in the superficial dermis. The serum level of IL-35 in the model group was lower than that in the control group (P <0.05). The serum levels of IL-4, IL-17 in the model group were higher than those in the control group (P <0.05). There was a negative correlation between IL-35 and IL-17 levels (P<0.05). The expressions of IL-12p35 and EBI3 mRNA were lower than those in the control group (P <0.05). Conclusion: The expression levels of IL-35 may play a role in the pathogenesis of atopic dermatitis.
