亚油酸对角质形成细胞屏障相关蛋白表达的影响

Effect of linoleic acid on the expression of keratinocytes barrier-associated protein

摘要: 目的:检测亚油酸对角质形成细胞屏障相关蛋白Filaggrin、keratin-14和claudin-1表达的影响。方法:选用不同浓度的亚油酸(2 ng/mL、10 ng/mL、20 ng/mL)刺激人角质形成细胞,细胞计数试剂盒(CCK-8)检测细胞存活率,并绘制细胞生存/抑制曲线;qRT-PCR和Western blot分别检测不同浓度亚油酸刺激后角质形成细胞Filaggrin、keratin-14和claudin-1的mRNA及蛋白水平的变化。结果:角质形成细胞生存率随亚油酸样品浓度升高而降低。角质形成细胞中Filaggrin和keratin-14的表达随着亚油酸样品浓度升高表达明显增加。claudin-1表达在2 ng/mL和10 ng/mL亚油酸作用下明显增加,而在20 ng/mL浓度下,claudin-1表达下降。结论:在2 ng/mL和10 ng/mL亚油酸作用下,Filaggrin、keratin-14和claudin-1表达随着亚油酸作用浓度升高而增加。

Abstract: Objective: To determine the effects of linoleic acid on the expression of skin barrier function-related proteins including Filaggrin, keratin-14 and claudin-1. Methods: Human keratinocytes were stimulated with different concentrations of linoleic acid (2, 10, 20 ng/mL). Cell viability was measured by cell counting kit (CCK-8), and cell survival/inhibition curves were drawn. qRT-PCR and Western blot were used to detect the mRNA and protein levels of Filaggrin, keratin-14 and claudin-1. Results: The survival rate of keratinocytes decreased with the increase of linoleic acid concentration. The expression of filaggrin and keratin in keratinocytes increased as the increase of linoleic acid concentration. The expression of claudin-1 was significantly increased at the concentration of 2 ng/mL and 10 ng/mL of linoleic acid, while decreased at 20 ng/mL. Conclusion: The expression of Filaggrin, keratin-14 increased with the concentration of linoleic acid at the concentration of 2 ng/mL and 10 ng/mL.
