扁平苔藓皮损中microRNA-155的表达及与Th17/Treg失衡的相关性

摘要: 目的:检测microRNA-155(miR-155)在皮肤扁平苔藓(CLP)中的表达,明确其与辅助性T细胞17 /调节性T细胞(Th17/Treg)失衡的相关性。方法:应用TaqMan探针法检测33例CLP患者和25名健康对照者外周血中miR-155,孤儿受体-γt(RORγt),白细胞介素-17A(IL-17A),叉状头/翼状螺旋转录因子P3(Foxp3)和IL-10的基因表达水平。结果:CLP中miR-155,RORγt、Foxp3和IL-17A 的表达较对照组显著升高,而IL-10的表达显著降低(均P<0.05);CLP组中,miR-155分别与RORγt、Foxp3和IL-17A的含量显著正相关(均P<0.001),与IL-10的含量无明显相关性(P>0.05)。结论:CLP中的miR-155过表达可能与Th17/Treg失衡有关。

Abstract: Objective: To detect the expression level of microRNA-155 (miR-155) and to determine the relationship with the imbalance of T-helper 17 cell/regulatory T cell (Th17/Treg) in the patients with cutaneous lichen planus (CLP). Methods: The expression of miR-155, retinoid-acid receptor-related orphan receptor gamma (RORγt), interleukin-17A (IL-17A), forkhead box P3 (Foxp3) and IL-10 in peripheral blood of 33 patients with CLP and 25 normal controls were detected by TaqMan probe. Results: Compared with the normal controls, the expression levels of miR-155, RORγt, Foxp3 and IL-17A were higher and the level of IL-10 expression was lower in the CLP patients with CLP (P<0.05). The level of miR-155 expression was positively correlated with the expression levels of of RORγt, Foxp3 and IL-17A in the patients with CLP (Ps<0.001). There was no correlation between the expression level of miR-155 and IL-10. Conclusion: The miR-155-upregulation existed in the CLP patients may be associated with the imbalance of Th17/Treg.
