柠檬烯治疗大鼠接触性皮炎模型疗效评价

Evaluation of the therapeutic effect of limonene on a rat model contact dermatitis

摘要: 目的:评价柠檬烯对大鼠接触性皮炎的治疗效果。方法:建立大鼠的接触性皮炎模型,36只大鼠随机分为阴性对照组、接触性皮炎模型组、低柠檬烯剂量组(100 mg/kg)、中柠檬烯剂量组(200 mg/kg)、高柠檬烯剂量组(400 mg/kg)及阳性对照地塞米松组(0.41 mg/kg),每组6只,连续给药10天。给药前后进行皮损评分、组织病理学及血清IL-2、IL-4、IL-6和IFN-γ水平检测。结果:柠檬烯组大鼠皮损评分,表皮增生、毛细血管扩张以及炎性细胞浸润及血清IL-2、IL-4、IL-6和IFN-γ水平均明显低于模型组,且高剂量柠檬烯最显著。柠檬烯高剂量组皮损改善和炎性指标改变情况优于地塞米松组(P<0.05)。结论:柠檬烯可明显改善接触性皮炎皮损并降低炎性因子水平。

Abstract: Objective: Investigate the therapeutic effects of limonene on contact dermatitis in rats. Methods: Establish a rat model of contact dermatitis, 36 rats were randomly divided into 6 groups with 6 rats in each group: blank group, contact dermatitis model group, low-dose limonene group (100 mg/kg), the medium-dose limonene group (200 mg/kg), the high-dose limonene group (400 mg/kg), and the positive control dexamethasone group (0.41 mg/kg). All groups were treated continuously for 10 days. Before and after treatment, lesion scoring, histopathological examination, and detection of serum levels of IL-2, IL-4, IL-6 and IFN-γ were performed. Results: Compared with the model group, the limonene-treated groups showed significantly lower lesion scores, reduced epidermal hyperplasia, capillary dilation, inflammatory cell infiltration, as well as decreased serum levels of IL-2, IL-4, IL-6 and IFN-γ, with the high-dose limonene group exhibiting the most prominent effect. Moreover, the high-dose limonene group showed superior improvements in skin lesions and inflammatory indicators compared with the dexamethasone group (P<0.05). Conclusion: Limonene can significantly ameliorate lesions of contact dermatitis and reduce the levels of inflammatory factors.
