灰树花多糖对TNF-α/IFN-γ诱导的HaCaT细胞炎症因子分泌的影响

Effect of Grifola frondosa polysaccharide on the inflammatory factors in HaCaT cells induced by TNF-α/IFN-γ

摘要: 目的:研究灰树花多糖(GFP)对TNF-α/IFN-γ诱导的HaCaT细胞炎症因子分泌的影响。方法:采用TNF-α/IFN-γ诱导HaCaT细胞炎症模型,以不同浓度的GFP(0.2 mg/mL、0.4 mg/mL和0.8 mg/mL)进行干预。分别采用qPCR和ELISA技术检测HaCaT细胞TARC、MDC、IL?6 和TNF-α mRNA及蛋白的表达水平,采用Western blot技术检测NF-κB和p-NF-κB表达水平。结果:GFP对HaCaT细胞炎性模型中TARC、MDC mRNA和蛋白及 p-NF-κB的 表达均有明显的抑制作用。结论:GFP具有明显的抗炎作用,其机制可能与调节NF-κB P65信号通路有关。

Abstract: Objective: To evaluate the effect of Grifola frondosa polysaccharide (GFP) on the inflammatory factors of HaCaT cells induced by TNF-α / IFN-γ. Methods: The HaCaT cells induced by TNF-α / IFN-γ were used as the model of inflammation. After 24h of intervention, the expression of TARC, MDC, IL-6 and TNF-α mRNA,and protein in HaCaT cells were detected by qPCR and ELISA respectively. The expression of NF-κB and p-NF-κB were detected by Western blot. Results: The mRNA and protein expression of TARC, MDC and p-NF-κB were inhibited after intervened by GFP. Conclusion: GFP has the anti-inflammatory effect, which may be associated with the NF-κB P65 signaling pathway.
