皮肤鳞状细胞癌AIM2表达水平与铁死亡指标的相关性研究

摘要: 目的:明确AIM2在皮肤鳞癌细胞中的表达及其与铁死亡的相关机制。方法:选取我院25例皮肤鳞癌组织及癌旁组织,免疫组化检测AIM2表达情况;CCK8实验检测AIM2对A431鳞癌细胞系的增殖影响,Western blot及qPCR方法检测铁死亡相关氧化还原调控因子HMOX1表达水平,抗氧化代谢物GSH、MDA等相关指标,同时电镜检测铁死亡细胞形态学变化。结果:AIM2在皮肤鳞癌细胞中表达上调,且与Ki-67高表达区域高度重叠;下调AIM2表达可以抑制A431细胞增殖,上调HMOX1表达,升高MDA水平、降低GSH水平,并诱导线粒体萎缩、嵴减少等铁死亡特征性改变。结论:AIM2在皮肤鳞癌细胞中高表达,通过抑制HMOX1表达降低铁死亡相关的脂质过氧化水平,促进肿瘤细胞增殖与疾病进展。

Abstract: Objective: To investigate the expression of AIM2 in cutaneous squamous cell carcinoma (cSCC) and its correlation with ferroptosis-related indicators. Methods: Single-center samples of 25 cases of cutaneous squamous cell carcinoma tissue and corresponding adjacent tissues were selected. AIM2 expression was detected by immunohistochemistry (IHC). The effect of AIM2 on the proliferation of A431 squamous cell carcinoma cell line was assessed using the CCK-8 assay. Western blot and qPCR were employed to measure the expression levels of the ferroptosis-related redox regulatory factor HMOX1, as well as related indicators such as the antioxidant metabolite GSH and MDA. Concurrently, transmission electron microscopy was used to observe the morphological changes associated with ferroptosis in cells. Results: AIM2 was upregulated in cSCC and highly overlapped with regions of high Ki-67 expression. Knockdown of AIM2 inhibited the proliferation of A431 cells, upregulated HMOX1 expression, increased MDA levels, decreased GSH levels, and induced ferroptosisassociated morphological changes including mitochondrial atrophy and reduced cristae. Conclusion: AIM2 is highly expressed in cSCC and promotes tumor cell proliferation and disease progression by suppressing HMOX1 expression and reducing ferroptosisrelated lipid peroxidation.
