氧化苦参碱对皮肤鳞状细胞癌Colo-16细胞株增殖及对EGFR表达的影响

Effects of oxymatrine on the proliferation of cutaneous squamous cell carcinoma Colo-16 cell and the expression of EGFR

摘要: 目的:明确氧化苦参碱对皮肤鳞状细胞癌(cSCC)Colo-16细胞株增殖及对EGFR表达的影响。方法:体外培养Colo-16细胞株,分为空白对照组、5%葡萄糖溶剂对照组、不同浓度氧化苦参碱(0.025 mg/mL、0.05 mg/mL、0.1 mg/mL、0.5 mg/mL、1 mg/mL、2 mg/mL)组。 MTT法测定各组Colo细胞株增殖情况,Western blot印迹法测定各组表皮生长因子受体(EGFR)及磷酸化表皮生长因子受体(pEGFR)的表达。结果:氧化苦参碱的各浓度组在24 h、48 h、72 h OD值均低于空白对照组及溶剂对照组,差异具有统计学意义(P<0.05)。氧化苦参碱各浓度组的OD值无显著差异(P>0.05)。不同浓度组与空白对照组相比及各不同浓度组之间,EGFR及磷酸化EGFR的表达差异均无统计学意义(P>0.05)。结论:氧化苦参碱在较低的浓度即有抑制Colo-16细胞株增殖的作用,而提高浓度不能增强抑制作用。氧化苦参碱并非通过抑制EGFR的表达来影响Colo-16细胞的增殖。

Abstract: Objective: To determine the effects of oxymatrine on the proliferation of cutaneous squamous cell carcinoma Colo-16 cell and the expression level of EGFR. Methods: The Colo-16 cells were cultured, and then were divided into the blank control group, 5% glucose solvents control group and different concentration of oxymatrine groups (0.025 mg/mL, 0.05 mg/mL, 0.1 mg/mL, 0.5 mg/mL, 1 mg/mL and 2 mg/mL). The proliferative activity of Colo-16 cell and the expression level of EGFR and phosphorylate EGFR were detected by MTT and Western blot respectively. Results: The optical density in the different concentration of oxymatrine were lower than those in the blank control group and 5% glucose solvents control group at 24 h, 48 h and 72 h, with significant differences (P<0.05). However, there was no significant difference of optical density between the different concentration of oxymatrine groups (P>0.05). There was no significant difference in the levels of EGFR and phosphorylate EGFR between all the groups (P>0.05).Conclusion: Oxymatrine can inhibit the proliferative activity of Colo-16 cell at lower concentration, in a non-concentration-dependent manner. Therefore, the effect of oxymatrine on the proliferative activity is not through inhibition of the expression of EGFR.
